Title: Block subdomain

I know I can configure my dir855 to block full websites. But is it also possible to block just some subdomains of a website?

You list the most inclusive domains such as example.com to include all of foo.example.com and bar.example.com.  It should then allow you to create a deny policy for foo.example.com only, allowing anything not specifically listed, such as bar.example.com and everything else at example.com.

I don't really understand what and where I need to configure this. Can you please explain a bit more?

Under Advanced->Access Control.  Enable Access Control.  Then add a Policy.  This specifies the systems, and times when the systems, are to be restricted.  Add the IPs or MAC Addresses of the machines to be controlled.

Then, under Advanced->Website Filter, add the specific domains you want blocked (or allowed, note the filter pulldown).
