Is it possible with the DIR-601 to have one of the IP's on the LAN route all traffic through a proxy server? I would like to have my Roku device (which I've setup as a static on the LAN side) to always connect though a proxy, while all my other devices connect to the internet normally.

I don't think so since these routers don't have any sort fo proxy management other than Port Forwarding and Virtual Server. If the proxy server is located on the WAN side, you might try setting up Virtual server and see if that might help with the Proxy, however I think you'd need to know port numbers and IP addresses for the proxy.

Is it possible to have an internal IP route via VPN to the internet with this router?

Most of these routers are pass thru for VPN so not alot of configuration is needed. However for some advanced VPNs, I think some can use the Virtual Server to set up and configure. I don't have any personal experience with these. You might check in FAQ Library (http://forums.dlink.com/index.php?board=304.0) for any articles saved regarding VS.
