Anyway to use the HOSTS file?

I have a rather extensive HOSTS file to block ads and such, and I'm wondering if it's possible to make the router use that file to block those same sites from any computer accessing the internet.

The reason I ask is because of my Wii, I'd like to block ad sites on it in the same way, but it doesn't use a HOSTS file to the best of my knowledge.

Yes (sort of), on my DIR-615C, it's under Advanced -> Website Filter. See the help page.

Have you considered using openDNS.org? It's a free DNS service that you point your router to for DNS lookups. It has similar features for blocking domains. For example, I setup openDNS to block doubleclick.net.

that only has limited entries. my HOSTS file has like 25,000 lines.

how do you tell the router to use opendns?

yes, it's free. You have to key in each domain to be blocked. I don't know if you can upload a file or what is the max number of blocked domains supported. YMMV.
